Human DNA Vaccines have gone a long way in the 20 years since their inception, but they still haven't shown to be a viable alternative to traditional protein or carbohydrate-based human vaccinations. While safety concerns were initially a deterrent, Human DNA Vaccines' Achilles heel is their low immunogenicity when compared to protein vaccinations. To improve DNA vaccine immunogenicity, a range of ways have been devised, including codon optimization, genetic adjuvants, electroporation, and complicated prime-boost regimens, each with its own set of benefits and drawbacks.
