Cytokines are produced signaling proteins that play important role in the initiation maintenance and resolution of immune responses. However the unique ability of cytokines to manage immune functions has garnered clinical interest in the context of cancer, autoimmunity, and infectious disease, but the use of cytokines-based therapeutics has been limited. Introduction


Cytokines go back to the times when interferons were first time discovered. Cytokines are pleiotropic proteins or little glycoprotein with a molecular weight is less than 29kDa cytokines are produced by several cell types like leukocyte which regulates immunity, inflammation, and hematopoiesis. About two hundred cytokines are recognized to date. They have a wide degree of helical structure and the molecular shares a similar polypeptide fold with four helical bundles. These are categorized on the basis from which they produce either Th1 cells or Th2 cells.


Cytokine Receptors


The cytokines use various biological effects through receptors present on the membranes of the responsive aimed cell. These receptors have an extracellular domain, a cytoplasmatic domain, and an individual membrane-spanning domain. There are two presences of polypeptide chains, one is the cytokine-specific α subunits and the other is a signal transducing β subunit.


The nature of the target cell to which the cytokine bind is determined by the presence of a specific membrane receptor. Cytokines and their receptor reveals a high affinity for each other and possess dissociation constants and for this reason high affinity, biological effects are produced by cytokines in picomolar concentration.


Role of Cytokines in Diseases


1. Rheumatoid Arthritis (RA)

It is a chronic autoimmune disease presenting stiffness, pain, and symmetrical synovitis of diarthrodial joints. Several cytokines are included in early events in the rheumatoid synovium. The mechanism of rheumatoid synovitis remains unclear, however, both antigen-specific and non-antigen-specific mechanisms may be involved.

So the cytokines are involved in the next level of rheumatoid synovitis which can be characterized by the migrations of immune and inflammatory cells from the blood to synovial space and tissue.


2. Cytokines in Brain

Numerous cytokines and their receptors are constitutively expressed and produced in the general human brain and their levels can differ in astrocytes and microglia. But in rodents, the maximum densities of IL-1, IL-2, IL-6, and TNF-α receptor was detected in the hippocampus and hypothalamus.


Cytokines As Therapeutic Targets


IL-1 AND IL-2 demonstrated potential as natural innuendo-stimulants to combat the immune deficiency of AIDS. clinical and experimental research supported the hypothesis that immune suppression of cancer and AIDS. familiar effects were proposed for TNF α for cancer patients. But the disturbing inflammatory response of cytokines like IL-1, IL-2, IL-3, IL-4, IL-6, IL-12, or TNFα in humans results in the sidelining of such therapy.

The cytokines are ubiquitous molecules having the function as key messengers for immune cells to maintain the right balance in the immune system. Their biological processes have a close effect on embryonic development, disease pathogenesis, non-specific response to infection, specific responses to the antigen, and others.
