FFPE samples-Types, Uses and Preparation

Formalin-fixed paraffin-embedded tissue specimens, often called FFPE samples, are a fantastic material for understanding human biology and disease pathways. It is widely used in clinical diagnosis and biological and medical research, especially in cancer and rare disease research. Due to the worldwide acceptance of in-vitro research, FFPE sample studies are critical for assessing morphology, molecular characteristics, and protein structure in human tissues. Furthermore, in contemporary medical research, using FFPE tissues for molecular pathological detection, such as tumor-related gene mutations and molecular detection of targeted drugs, has progressed rapidly from scientific research to in-vitro pathological diagnosis. Furthermore, retrospective gene research is becoming more popular in tumor molecular biology. However, because most tissue samples used in scientific research are FFPE tissue blocks and FFPE tissue sections, the amount of DNA and RNA extracted from them directly impacts the outcome of molecular experiments.

How are FFPE samples prepared?

FFPE is a subset of the human biospecimen preservation method that has been in use for many years. Collecting tissue specimens from disease-state or healthy human donors is the first step in preparing FFPE samples. The extraction of specimens is mainly done during biopsies and clinical and surgical procedures by a trained medical doctor or surgeon. The excised human samples are immersed in 10% neutral buffered formalin for at least 24 hours. This process is known as formalin fixation. Later, the fixed samples are dehydrated with ethanol concentrates. The tissue is then embedded in paraffin wax to create FFPE blocks, which preserve the proteins and tissue structures. Finally, these FFPE blocks are sectioned into specific sizes to prepare FFPE specimens that are investigated and authorized by a certified histopathologist for research use. FFPE samples in the form of FFPE blocks, curls, and slides can be stored for long years without degradation. Moreover, FFPE is stored at room temperature and thus does not require highly sophisticated and expensive tissue preservation units.
